| Plasma Abundant Proteins |

Plasma proteins can be roughly divided into Highly-Abundant Proteins (HAP), Moderately-Abundant Proteins (MAP) and Low-Abundant Proteins (LAP). The majority of the clinically relevant biomarkers reside within the low-abundant plasma protein fraction. Biomarker discovery requires removal of HAP since these proteins block the low-abundant protein fraction, where many biomarkers reside. The HAP fraction constitutes over 96% of the total protein mass. It includes such proteins as Human Albumin (HSA), IgG, Fibrinogen, Transferrin and many other proteins. For novel biomarker discovery and validation, it is useful to remove HAP and further partition MAP and LAP.
GenWay specializes in immunoaffinity fractionation by applying its IgY microbead system known as Seppro®. Based on the advantages of IgY antibodies, GenWay has developed various types of IgY immunoaffinity columns for removing HAP or fractionating MAP from LAP. Numerous studies have demonstrated that IgY immunoaffinity fractionation of plasma or serum is highly specific and reproducible. |

| IgY-14 /SuperMix fractionation |
| IgY-14 captures the top14 abundant human plasma proteins. Human Serum Albumin, IgG, IgA, IgM, Fibrinogen, Transferrin, Haptoglobin, α2-Macroglobulin, α1-Acid Glycoprotein, α1-Antitrypsin, C3 complement, HDL (Apo A-I & Apo A-II), and LDL (ApoB). SuperMix is an IgY-affinity column that enables one-step specific separation for identification of hundreds of MAP and LAP. In a Case Study on a sample of 250 µl, using GenWay’s SuperMix column coupled with SDS-PAGE and LC/MS-MS, within 7 days, total 923 proteins in two fractions were identified with high confidence. |

| Specific needs for fractionation
of plasma proteins can be met by customizing mixture of different
IgY microbeads against different target proteins. For example, anti-BSA
IgY antibodies can be used to mixed with other IgY microbeads for
making IgYC12 for cow plasma fractionation. |

The IgY microbeads can also be used individually for specifically removing or analyzing target proteins and physiologically associating biomolecules. Additional IgYs against abundant plasma/serum, other body fluid or cellular proteins can be readily produced to meet your needs.
The convenient feature of this service is that variable volumes of samples (10 µl to 1 ml or more) can be readily arranged for processing according to the client’s needs. Generally the services are completed within one week after samples are received from customers.
The service package includes designing the process with clients, obtaining
samples, conducting the immunoaffinity partitioning process, and delivering
the processed flow-through and eluted samples back to the client. |
