Details for MAST CELL CHYMASE, Antibody

Specificity: MAST CELL CHYMASE

NCBI Gene ID: 1215

Isotype: Specificity: MAST CELL CHYMASE

Clone: CC1 Immunogen: Purified human skin chymase

Fusion Partner: Spleen cells from immunised BALB/c mice were fused with cells of the P3/NSI/I.Ag4.1 mouse myeloma cell line.

Specificity Note: This product binds to human mast cell chymase, an important marker of mast cells and also an important mediator of inflammation.

This product may be used to detect mast cells in routinely fixed tissues, and has been used to study the distribution of mast cells in skin, synovium, lung and heart.

Binding of This product to chymase is not reduced in the presence of heparin, suggesting that the antibody does not bind to the heparin-binding domain of chymase. Immunohistology: This product does not require protein digestion pre-treatment of paraffin sections. This product does not require antigen retrieval using heat treatment prior to staining of paraffin sections.

Histology: Skin

Preparation: Purified IgG prepared by affinity chromatography on Protein G from tissue culture supernatant

Buffer Solution: Phosphate buffered saline pH7.4

Preservative Stabilisers: 0.09%Sodium Azide

Suggested Dilution: Suggested Dilution: Immunohistology - Paraffin - 1/500 - 1/1000 Function: Major secreted protease of mast cells with suspected roles in vasoactive peptide generation, extracellular matrix degradation, and regulation of gland secretion.

Catalytic Activity: Preferential cleavage: Phe-|-Xaa > Tyr-|-Xaa > Trp-|-Xaa > Leu-|-Xaa.

Subcellular Location: Secreted. Cytoplasmic granule. Note=Mast cell granules.

Tissue Specificity: Mast cells in lung, heart, skin and placenta.

Similarity: Belongs to the peptidase S1 family. Granzyme subfamily [view classification].

Similarity: Contains 1 peptidase S1 domain [view classification]. 1. Buckley. M.G. et al. (1999) The detection of mast cell subpopulations in formalin-fixed human tissues using a new monoclonal antibody specific for chymase.
