Specificity: M2I-4 reacts with an internal epitope of MRP2, a 190-200 kD transmembrane protein earlier known as the canalicular multi-organic anion transporter cMOAT, absent in patients with the Dubin-Johnson syndrome, an autosomal recessive liver disorder characterized by chronic conjugated hyperbilirubinemia. MRP2 is a member of the MRP family of multidrug resistance related proteins, and MRP2 overexpression has been observed in a subset of cisplatin resistant cell lines. M2I-4 was raised against a bacterial fusion protein of MRP2, containing amino acids 215-310 of the protein. M2I-4 did not cross-react with the human MDR1, MRP1, MRP3 and MRP5 gene products
Mab Producing Cells: The hybridoma cell line was obtained by fusion of lymph node cells from an immunized mouse (Balb/c) with SP2/O mouse myeloma cells
Uses: M2I-4 has potential value for detection of MRP2-mediated drug-resistance in human tumor samples. Immunocytochemistry: use 1:20-50 dil. on acetone fixed cytospin preparations. For immunohistochemistry: M2I-4 (use 1:20) on acetone fixed frozen sections can be followed by incubation with rabbit anti- mouse IgG (1:25, Dako) and a monoclonal mouse APAAP complex (1:50, Dako). After incubation of M2I-4 (use 1:20) and washing, slides can be incubated with biotinylated rabbit anti-mouse IgG (1:100, Jackson, West Grove) and streptavidin conjugated to horseradish peroxidase (1:500, Zymed, San Francisco, CA). Flow cytometry: optimal conditions still to be established. Western blotting: use 1:20-50 dil, and anti-mouse-HRP
Culture Medium: IMDM (BioWhittaker), supplemented with Nutridoma-SP (Boehringer, Indianapolis, USA), without serum or added enzymes. Antibody containing supernatant has been concentrated and filtered through a 0.22 micron filter

Function: Mediates hepatobiliary excretion of numerous organic anions. May function as a cellular cisplatin transporter.
Subcellular Location: Membrane; Multi-pass membrane protein.
Tissue Specificity: Found on the apical membrane of polarized cells in liver, kidney and intestine. The highest expression is found in liver.
Disease: Defects in ABCC2 are the cause of Dubin-Johnson syndrome (DJS) [MIM:237500]. DJS is an autosomal recessive disorder characterized by conjugated hyperbilirubinemia, an increase in the urinary excretion of coproporphyrin isomer I, deposition of melanin-like pigment in hepatocytes, and prolonged retention of sulfobromophthalein, but otherwise normal liver function.
