The Mannose-6 Phosphate Receptor (also known as Insulin-like Growth Factor 2 Receptor and CD222) is a type I transmembrane protein which is expressed ubiquitously. The majority of CD222 molecules are located intracellularly. CD222 is a 250 kDa transmembrane protein with multiple cellular functions. Its main functions are internalization of IGF-2. internalization or sorting of lysosomal enzymes and other M6P-containing proteins as well as regulation of TGF-beta activity. Furthermore it is crucial for the internalization and degradation of IGF-2 and thereby controls cell growth. A role of CD222 in regulation of fibrinolysis. cell adhesion and migration has been found .

Function: Transport of phosphorylated lysosomal enzymes from the Golgi complex and the cell surface to lysosomes. Lysosomal enzymes bearing phosphomannosyl residues bind specifically to mannose-6-phosphate receptors in the Golgi apparatus and the resulting receptor-ligand complex is transported to an acidic prelyosomal compartment where the low pH mediates the dissociation of the complex. This receptor also binds IGF2.

Subunit: Binds GGA1, GGA2 and GGA3.

Subcellular Location: Lysosome membrane; Single-pass type I membrane protein.

Domain: Contains 15 repeating units of approximately 147 AA. The most highly conserved region within the repeat consists of a stretch of 13 AA that contains cysteines at both ends.

Similarity: Belongs to the MRL1/IGF2R family.

Similarity: Contains 1 fibronectin type-II domain.
