 MPO ANTIBODY TARGET DESCRIPTION:
Synonym Names for MPO antibody: MPO; BIOTINYLATED MONOCLONAL ANTIBODY TO HUMAN MYELOPEROXIDASE (MPO); EC 1.11.1.7; MPO; Myeloperoxidase

The monoclonal antibody 266-6K1 reacts with Myeloperoxidase (MPO). a glycoprotein with an alpha2beta2 heteromultimer expressed in all cells of the myeloid linage. MPO is abundantly present in azurophilic granules of polymorphonuclear neutrophils. It is an important enzyme used during phagocytic lysis of engulfed foreign particles which takes part in the defense of the organism through production of hypochlorous acid (HOCl). a potent oxidant. MPO is rapidly released by activated polymorphonuclear neutrophils. Involvement of MPO has been described in numerous diseases such as atherosclerosis. lung cancer. Alzheimer's disease and multiple sclerosis. Autoimmune antibodies to MPO are involved in Wegeners disease. Since the discovery of MPO deficiency. initially regarded as rare and restricted to patients suffering from severe infections. MPO has attracted more clinical attention. In experimental studies antibodies to MPO can be used for various purposes ranging from flow cytometric analysis to detection of polymorphonuclear neutrophils in tissue sections.

Function: Part of the host defense system of polymorphonuclear leukocytes. It is responsible for microbicidal activity against a wide range of organisms. In the stimulated PMN, MPO catalyzes the production of hypohalous acids, primarily hypochlorous acid in physiologic situations, and other toxic intermediates that greatly enhance PMN microbicidal activity.

Catalytic Activity: Donor + H2O2 = oxidized donor + 2 H2O.

Catalytic Activity: Cl- + H2O2 = HOCl + 2 H2O.

Cofactor: Binds 1 calcium ion per heterodimer.

Cofactor: Binds 1 heme B (iron-protoporphyrin IX) group covalently per heterodimer.

Subunit: Tetramer of two light chains and two heavy chains.

Subcellular Location: Lysosome.

Disease: Defects in MPO are the cause of myeloperoxidase deficiency (MPD) [MIM:254600]. MPD is an autosomal recessive defect that results in disseminated candidiasis.

Similarity: Belongs to the peroxidase family. XPO subfamily.

Summary: Myeloperoxidase (MPO) is a heme protein synthesized during myeloid differentiation that constitutes the major component of neutrophil azurophilic granules. Produced as a single chain precursor, myeloperoxidase is subsequently cleaved into a light and heavy chain. The mature myeloperoxidase is a tetramer composed of 2 light chains and 2 heavy chains. This enzyme produces hypohalous acids central to the microbicidal activity of netrophils.

MYELOPEROXIDASE (MPO) reacts with human.
