Application Note: For IHC: Use at a dilution of 1:50-1:100 for 30mins at RT (ABC method). For formalin fixed paraffin embedded sections no special pretreatment is required. This antibody stains S100 protein in Schwannomas, ependymomas, astrogliomas, almost all benign and malignant melanomas and their metastases. Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

S100 belongs to the family of calcium binding proteins such as calmodulin and troponin C. S100A and S100B proteins are two members of the S100 family. S100A is composed of an alpha and beta chain whereas S100B is composed of two beta chains.

Function: Weakly binds calcium but binds zinc very tightly-distinct binding sites with different affinities exist for both ions on each monomer. Physiological concentrations of potassium ion antagonize the binding of both divalent cations, especially affecting high-affinity calcium-binding sites.

Subunit: Dimer of either two alpha chains, or two beta chains, or one alpha and one beta chain.

Subcellular Location: Cytoplasm.

Tissue Specificity: Highly prevalent in heart. Also found in lesser quantities in skeletal muscle and brain.

Similarity: Belongs to the S-100 family.

Similarity: Contains 2 EF-hand domains.
