c-AMP-dependent Protein Kinase (PKA) is a serine/threonine kinase that regulates a number of cellular processes including proliferation, ion transport and gene transcription. PKA is composed of conserved catalytic subunits and regulatory subunits that dissociate upon activation by cAMP. The catalytic subunit of PKA contains the activation loop and mediates DNA binding and substrate recognition. The catalytic subunit is assembled and expressed as an active form and is phosphorylated on threonine 197 by PDK-1 in the activation loop. Phosphorylation of threonine 197 is critical for PKA maturation, optimal conformation and catalytic activity and plays a key role in mediating its biological functions.
Function: Phosphorylates a large number of substrates in the cytoplasm and the nucleus.
Catalytic Activity: ATP + a protein = ADP + a phosphoprotein.
Enzyme Regulation: Activated by cAMP.
Subunit: A number of inactive tetrameric holoenzymes are produced by the combination of homo- or heterodimers of the different regulatory subunits associated with two catalytic subunits. cAMP causes the dissociation of the inactive holoenzyme into a dimer of regulatory subunits bound to four cAMP and two free monomeric catalytic subunits.
Subcellular Location: Cytoplasm (By similarity). Nucleus (By similarity). Note=Translocates into the nucleus (monomeric catalytic subunit) (By similarity). The inactive holoenzyme is found in the cytoplasm (By similarity).
Tissue Specificity: Isoform 2 is sperm specific.
Ptm: Asn-3 is partially deaminated to Asp giving rise to 2 major isoelectric variants, called CB and CA respectively (By similarity).
Similarity: Belongs to the protein kinase superfamily. AGC Ser/Thr protein kinase family. cAMP subfamily.
