 Application Note: Dot: Use at an assay dependent dilution. ELISA: 1:1,000 - 1:5,000. IHC-P: 1:200. Quench peroxidases with methanol and H2O2 3% for 15 min. Perform heat mediated antigen retrieval via the microwave method (3x5 min).before commencing with IHC staining protocol. Incubate antibody overnight. A modified streptavidin technique was used (Kwik kit) (see Koukourakis MI et al reference). IM: Use at an assay dependent dilution. WB: 1:1,000-1:10,000. Please note: The non-conjugated version of this antibody (18-272-197038) has been used in WB in the reference Zaman K et al (protocol available on the datasheet for 18-272-197038). Not yet tested in other applications.Optimal dilutions/concentrations should be determined by the end user.

Lactate dehydrogenase (LDH) is present in a wide variety of organisms, including plants and animals. It is an oxidoreductase which catalyses the interconversion of pyruvate and lactate with concomitant interconversion of NADH and NAD+. As it can also catalyze the oxidation of hydroxybutyrate, it is occasionally called Hydroxybutyrate Dehydrogenase (HBD). There are 5 different isoenzymes of LDH, LDH-1 to LDH-5, each composed of 4 subunits which may be of 2 different types - M and H subunits. These subunits are encoded by two different genes: The M subunit is encoded by gene LDHA whilst the H subunit is encoded by LDHB. Usually LDH-2 is the predominant form in the serum. An LDH-1 level higher than the LDH-2 level suggests myocardial infarction (damage to heart tissues releases heart LDH, which is rich in LDH-1, into the bloodstream).

Catalytic Activity: (S)-lactate + NAD+ = pyruvate + NADH.

Pathway: Fermentation; pyruvate fermentation to lactate; (S)-lactate from pyruvate: step 1/1.

Subunit: Homotetramer.

Subcellular Location: Cytoplasm.

Disease: Defects in LDHB are a cause of hereditary LDHB deficiency [MIM:150100]. LDHB deficiency is usually asymptomatic.

Similarity: Belongs to the LDH/MDH superfamily. LDH family.
