 Specificity: The antibody reacts with the extra-cellular part of the TNF-R II and with the soluble receptor. TNF-R II is present on most cell types and is considered to play a role in cell stimulation by TNF-alpha. TNF-R II molecule is shown to be responsible for stimulation of activated T-lymphocytes by TNF-alpha.

 Application Note: The antibody is an agonistic antibody useful for detection of TNF-R II. It can be used for immunoassays, Western blotting, flow cytometry and immunoprecipitation. The reactivity of the antibody with cellbound or soluble TNF-Receptor is not inhibited by high concentrations of TNF-a. Minimal crossreactivity is present with human TNF-a. Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user.

TNF-R II is present on most cell types and is considered to play a role in cell stimulation by TNF-a. TNF-R II molecule is shown to be responsible for stimulation of activated T-lymphocytes by TNF-a.

Function: Receptor with high affinity for TNFSF2/TNF-alpha and approximately 5-fold lower affinity for homotrimeric TNFSF1/lymphotoxin-alpha. The TRAF1/TRAF2 complex recruits the apoptotic suppressors BIRC2 and BIRC3 to TNFRSF1B/TNFR2. This receptor mediates most of the metabolic effects of TNF-alpha. Isoform 2 blocks TNF-alpha-induced apoptosis, which suggests that it regulates TNF-alpha function by antagonizing its biological activity.

Subunit: Binds to TRAF2.

Subcellular Location: Isoform 1: Cell membrane; Single-pass type I membrane protein.

Subcellular Location: Tumor necrosis factor-binding protein 2: Secreted.

Ptm: Phosphorylated; mainly on serine residues and with a very low level on threonine residues.

Ptm: A soluble form (tumor necrosis factor binding protein 2) is produced from the membrane form by proteolytic processing.

Pharmaceutical: Available under the name Enbrel (Immunex and Wyeth-Ayerst). Used to treat moderate to severe rheumatoid arthritis (RA). Enbrel consist of the extracellular ligand-binding portion of TNFRSF1B linked to an immunoglobulin Fc chain. It binds to TNF-alpha and blocks its interactions with receptors.

Similarity: Contains 4 TNFR-Cys repeats.

TNF receptor II reacts with human - not yet tested in other species.
