Applications Notes : Suitable for use in a variety of gel techniques including radial immunodiffusion (RID), double diffusion, and immunoelectrophoresis (IEP). The use of 3% PEG 6000 with 1.2% agarose in a suitable buffer (such as Tris borate EDTA or Tris-barbital pH >8.2) is recommended. Suitability for use in other techniques such as rocket IEP, immunohistochemical procedures, nephelometry, enzyme-linked immunosorbent assays and Western blot has not been assessed but use in such assays should not necessarily be excluded. Sheep anti Alpha 1-Antitrypsin. Sheep Antibody to Human Alpha-1-Antitrypsin

Function: Inhibitor of serine proteases. Its primary target is elastase, but it also has a moderate affinity for plasmin and thrombin.

Subcellular Location: Secreted.

Domain: The reactive center loop (RCL) extends out from the body of the protein and directs binding to the target protease. The protease cleaves the serpin at the reactive site within the RCL, establishing a covalent linkage between the carboxyl group of the serpin reactive site and the serine hydroxyl of the protease. The resulting inactive serpin-protease complex is highly stable (By similarity). Variability within the reactive center loop (RCL) sequences of Serpina1-related genes may determine target protease specificity.

Miscellaneous: Murine alpha-1-antitrypsin is represented by a cluster of up to 6 individual Serpina1-related genes. The precise complement of Serpina1-related genes present varies according to the strain of the animal.
