 Experiment Name: Immunoblot analysis for Enolase 1 in U87 cells after irradiation (7 Gy).

 Experiment Background: To determine whether the radiation-induced change in a gene's translational activity (as indicated by polysome binding) corresponds with a change in its protein product (here Enolase 1),

 Experiment Steps: 1. Rinse cells with ice-cold PBS.
2. Scrap cells into lysis buffer containing 50 mmol/L Tris (pH 7.4), 150 mmol/L NaCl, 1 mmol/L EDTA, 1% NP40, supplemented with protease inhibitor cocktail and phosphatase inhibitors I and II.
3. Separate insoluble material after centrifugation at 14,000 rpm.
4. Subject proteins to SDS-PAGE using NuPage 4% to 12% gels and NuPage MES or MOPS buffers.
5. After electrophoresis, electroblot gels onto polyvinylidene difluoride membranes.
6. At room temperature block the nonspecific sites on the membranes for 30 minutes with 5% nonfat milk in TBS supplemented with 0.2% Tween 20 (TBS-T).
7. Block membranes in blocking solution overnight at 4C with enolase 1.
8. Wash membranes thrice in TBS-T.
9. Incubate with the appropriate horseradish peroxidase-conjugated secondary antibody at a 1:2,000 dilution in blocking solution for 1 hour at room temperature.
10. Again wash membranes thrice in TBS-T.
11. Develop membranes with enhanced chemiluminescence western blotting detection reagents.
12. Visualize Membranes with a Typhoon scanner.

Synonym Names for ENO1 antibody: ENO1; ENO1L1, MBP-1, MPB1, NNE, PPH; EC 4.2.1.11; Enolase 1; Enolase

ENO1 is one of three enolase isoenzymes found in mammals; it encodes alpha-enolase, a homodimeric soluble enzyme, and also encodes a shorter monomeric structural lens protein, tau-crystallin. The two proteins are made from the same message. The full length protein, the isoenzyme, is found in the cytoplasm. The shorter protein is produced from an alternative translation start, is localized to the nucleus, and has been found to bind to an element in the c-myc promoter. A pseudogene has been identified that is located on the other arm of the same chromosome.

Catalytic Activity: 2-phospho-D-glycerate = phosphoenolpyruvate + H2O.

Cofactor: Magnesium. Required for catalysis and for stabilizing the dimer (By similarity).

Pathway: Carbohydrate degradation; glycolysis; pyruvate from D-glyceraldehyde 3-phosphate: step 4/5.

Similarity: Belongs to the enolase family.
