Application Info ELISA Capture: Purified JES3-9D7 monoclonal antibody is the preferred capture antibody in a
sandwich ELISA for quantifying human and/or viral IL-10 proteins.1-5 Purified JES3-9D7 antibody should
be paired with biotinylated JES3-12G8 antibody (Aviva_CatNum. GWB-85F05B) as the detection antibody, with
purified recombinant human IL-10 as the standard.
Immunofluorescence/Flow Cytometry: The JES3-9D7 antibody is useful for intracytoplasmic staining
and flow cytometric analysis to identify and enumerate IL-10-positive cells within mixed cell populations.
Recombinant human IL-10 or unlabeled Rat IgG1 should be used as a specificity control.
Western Blotting: The purified JES3-9D7 antibody is also useful for Western blotting applications. A
concentration of 1-5 μg/mL (in conjunction with AP-labeled Goat Anti-Rat-Ig, Aviva_CatNum. GWB-52F78C) has been
found to enable detection of <= 100 ng/lane of recombinant human IL-10 under reducing conditions.
Neutralization: The LE/AF form of JES3-9D7 can be used for neutralizing human/viral IL-10 bioactivity.
A suitable LE/AF Rat IgG1 (Aviva_CatNum. GWB-1D80D3) should be used as an isotype control.
Since applications vary, each investigator should determine the optimal concentration appropriate for individual applications.
