Product Name: Cytomegalovirus IgG - ELISA

# of Samples: 1 x 96 Assays

Intended Use: The Cytomegalovirus IgG ELISA is intended for the qualitative determination of IgG class antibodies against CMV in human serum or plasma (citrate).

Introduction: Cytomegalovirus (CMV) is a member of the herpesvirus group (Betasubfamily, DNA virus of 150-200 nm). These viruses share a characteristic ability to remain dormant within the body over a long period. Initial CMV infection, which may have few symptoms, is always followed by a prolonged, inapparent infection during which the virus resides in cells without causing detectable damage or clinical illness. Severe impairmant of the body´s immune system by medication or disease consistently reactivates the virus from the latent or dormant state.
CMV is found universally throughout all geographic locations and socioeconomic groups, and infects between 50% and 85% of adults. CMV infection is more widespread in developing countries and in areas of lower socioeconomic conditions. For the vast majority of people, CMV infection is not a serious problem, but it is to certain high-risk groups:
the unborn baby during pregnancy
people who work with children
immunocompromised persons, such as organ transplant recipients and persons infected with HIV
The presence of virus resp. infection may be identified by
Serology: CBR, Detection of antibodies by ELISA

Principles of the assay: The qualitative immunoenzymatic determination of IgG-class antibodies against CMV is based on the ELISA (Enzyme-linked Immunosorbent Assay) technique.
Microtiter strip wells are precoated with CMV antigens to bind corresponding antibodies of the specimen. After washing the wells to remove all unbound sample material horseradish peroxidase (HRP) labelled anti-human IgG conjugate is added. This conjugate binds to the captured CMV-specific antibodies. The immune complex formed by the bound conjugate is visualized by adding Tetramethylbenzidine (TMB) substrate which gives a blue reaction product.
The intensity of this product is proportional to the amount of CMV specific IgG antibodies in the specimen. Sulphuric acid is added to stop the reaction. This produces a yellow endpoint colour. Absorbance at 450 nm is read using an ELISA microwell plate reader.

Storage and Stability: The reagents are stable up to the expiry date when stored at 2...8 °C.

Limitations of the Test: Bacterial contamination or repeated freeze-thaw cycles of the specimen may affect the absorbance values. Diagnosis of an infectious disease should not be established on the basis of a single test result. A precise diagnosis should take into consideration clinical history, symptomatology as well as serological data. In immunocompromised patients and newborns serological data only have restricted value.
